10羽14日龄鸡分别以马立克氏病毒(MDV)BJ-1株接毒,采集羽毛根作为MDV琼扩抗原和MDVDNAdot-blot杂交的动态检测样品。接毒后第10d至第8周的检测结果显示,鸡羽毛根MDV琼扩抗原及MDVDNA可分别在接毒后第14d(37.5%,3/8)和第12d(33.3%,3/9)检出;接毒后第17d至第5周,鸡羽毛根MDV琼扩抗原的阳性检出率均大于80%;第6,7和8周时,则分别降至66.7%(4/6),50%(3/6)和40%(2/5)。而MDVDNA的检测,除1只鸡直到第4周才呈阳性外,其余在接毒后第14d至第8周中均保持100%的阳性检出率。
The 14oldday chickens were inoculated with Marek′s disease virus (MDV) strain Beijing1 (BJ1). During 10 days to 8 weeks postinjection (PI), a detection of MDV antigens and DNA in the feather tips from same injected chickens were made using two assays: agargelprecipitation (AGP) and dotblot DNA hybridization. The results showed MDV in the feather tips were identified 14 days PI (37.5%, 3/8) by AGP and 12 days PI (33.3%, 3/9) by DNAhybridization. Following detection, positive rates of MDV were always above 80% from 17 days to 5 weeks PI by AGP and were decreased significantly to 66.7% (4/6), 50%(3/6) and 40%(2/5) at week 6,7 and 8 PI by AGP, respectively. However, the exception of one bird was negative since week 4 PI. the positive rates were always 100% from 14 days to 8 weeks PI by DNAhybridization.
