摘要
目的探讨PTEN在乳腺癌中表达与Akt、Cyclin D1、MMP-2蛋白表达的相关性及临床意义。方法采用链霉菌抗生物素蛋白-过氧化酶法(S-P法)检测PTEN、Akt、Cyclin D1、MMP-2蛋白在65例乳腺导管癌和15例正常乳腺和纤维腺瘤组织中的表达。结果15例正常乳腺和纤维腺瘤组织中PTEN表达最高。65例乳腺导管癌中,淋巴转移32例,无淋巴转移33例,PTEN的表达率在乳腺导管原位癌和分化较好(Ⅰ、Ⅱ)的乳腺浸润性导管癌中表达率高,与Ⅲ、Ⅳ期乳腺浸润导管癌中PTEN表达有明显差异(P<0.01)。PTEN表达与Akt、Cyclin D1、MMP-2表达呈负相关。结论PTEN蛋白表达下降发生在乳腺癌的早期,并且与乳腺癌的发展预后密切相关。PTEN蛋白减少促使PI3K/Akt信号转导通路的活性可能是乳腺癌发生发展的分子机制,Cyclin D1、MMP-2参与了该过程。
Objective to investigate the relativity and clinic significance of expression of protein PTEN and expression of protein Akt, Cyclin D1, MMP-2 in breast cancer. Methods Streptomycete avidin-Peroxidase method(S-P)was used to examine expression of protein PTEN, Akt, Cyclin D1, MMP-2 in 65 cases of breast duct carcinoma and 15 cases of normal mammary gland and breast fibroadenoma. Results The exppression of PTEN in 15 cases of normal mammary gland and breast fibroadenoma tissue is the highest. Among 65 cases of breast ductal carcinoma(32 cases suffered from lymphatic metastasis and 33 cases didn't), expression of PTEN in ductal carcinoma in situ of the breast and well-differentiated (Ⅰ、Ⅱ) breast invasive ductal carcinoma is high in these cases. It exists a significant difference(P〈0.01) compared to exppression of PTEN in Ⅲ,Ⅳ phase breast invasive ductal carcinoma. Exppression of F1EN is negatively relative to exppression of Akt, Cyclin D1, MMP-2. Conclusions Depress of exppression of protein PTEN occurrs in early phase of breast cancer, and it is closely related to development and prognosis of breast cancer. Decrease of protein PTEN promotes activity of PI3K/Akt signal transduction pathway,which could be one of molecule mechanism of breast cancer growth and development. Cyclin D1, MMP-2 were involved in this process.
