基质金属蛋白酶及其组织抑制因子在肺癌中的表达及临床意义 被引量:1

Expresion of Matrix Metalloproteinase and Tissue Inhibitor in Lung Cancer and Their Clinical Significance

摘要 目的探讨基质金属蛋白酶MMP-2及其组织抑制因子-2与肺癌侵袭和转移的关系。方法选取肺癌住院手术患者共45例,其中男30例,女15例,鳞癌、腺癌、小细胞肺癌各15例。术后临床分期Ⅰ期15例,Ⅱ期20例,Ⅲ期10例。取上述肺癌患者切除标本距离肿瘤组织3cm以外的正常组织15例为组织对照组,采用免疫组化S-P法分别对肺癌患者组织中的MMP-2及TIMP-2进行测定,采用χ2检验进行统计学分析。结果肺癌组织中,MMP-2、TIMP-2组织表达明显高于对照组(P<0.01,P<0.05),在Ⅰ、Ⅱ、Ⅲ期中,MMP-2在组织中的表达随分期的升高而增加(P<0.05),但TIMP-2的表达则相反,随肿瘤分期增加而降低(P<0.05)。结论基质金属蛋白酶-2在肺癌侵袭和转移中具有重要的作用,可以用来评价肿瘤的恶性程度和估计预后。 Objective To explore the relationship of matrix metalloproteinase-2, tissue inhibitor matrix metalloproteinase-2 and infiltrition and metastasis in lung cancer. Methods Forth-five patients with lung cancer were involved in this study. All patients got operations after admitted to hospital. 30 were male and 15 were female. Pathohistological investigations revealed squmous cell carcinoma (n=15),adenocarcinoma (n=15)and small cell carcinoma(n=15). According to the clinical stage,15 cases were in stage Ⅰ,20 cases in stage Ⅱ and 10 cases in stage Ⅲ. Normal tissues 3cm distance away from the above carcinoma tissues were as control group. Paraffin-embedded tissue specimens were tested by immunohistochemistry (IHC)using S-P method. The results were analyzed acoording to the statistics by X^2 test. Results Expression rates of MMP-2 ,TIMP-2 in lung cancer group were significantly higher than those in the control group. MMP-2 expression increased with the histological stage while TIPM-2 decreased with the stage. Conclusion MMP-2 played an important role in evaluating tumor metastasis and might be used to supervise carcinoma.
