摘要
[目的]探讨不同培养条件对枸杞组培苗玻璃化的影响。[方法]以"宁杞3号"的幼嫩茎段为外植体,以MS为基本培养基,研究6-BA浓度、蔗糖浓度、琼脂浓度、培养温度、光照时间对枸杞组培苗玻璃化现象的影响。[结果]当6-BA浓度为0.2 mg/L、蔗糖浓度为3%、琼脂浓度为6.5 g/L、培养温度25℃、光照时间12 h/d(日光灯,光照强度2000 lx)时,枸杞组培苗繁殖系数最高,玻璃化率较低。[结论]综合调控枸杞组织培养条件,可以减少玻璃化现象的发生。
[ Objective] The study aimed to discuss the effects of different culture conditions on the vitrification of Lycium barbarum L. seedlings. [ Method] With the tender stems of Ningqi 3 as the explants and MS as basic medium, the effects of 6-BA concn., cane sugar cohen,, agar concn., culture temperature and illumination time on the vitrification of tissue culture seedlings of L. barbarum were studied. [Result]When 6-BA was 0.2 mg/L, sucrose was 3%, agar was 6.5 g/L, culture temperature is 25 ℃ and illumination time is 12 h/d resp., the tissue culture seedlings of L. barbarum had the highest bud multiplication coefficient and the lowest vitrification rate. [ Conclusion] Regulating the tissue culture conditions of L. barbarum comprehensively could reduce the occurenee of the vitrification phenomena.
