摘要
本文通过正交实验研究马来眼子菜多糖的提取工艺,并初步研究其组成性质。采用热水提取马来眼子菜多糖(Potamogeton malaianus polysaccharide PMPS),经乙醇沉淀,活性碳脱色,Sevage法去蛋白,DEAE-Cellulose离子交换和Sephadex G-25柱层析纯化,电泳鉴定纯度和纸层析分析其组成。确定最佳提取条件为加10倍体积的水,80℃提取3 h,所得二个多糖电泳均为单一组分,层析证明马来眼子菜多糖Ⅰ(PMPSⅠ)由D-果糖和D-木糖组成,马来眼子菜多糖Ⅱ(PMPSⅡ)则含有D-葡萄糖、D-果糖和另一未知单糖成分。
This paper studied on the isolation technology of polysaccharide from Potamogeton raalaianus by orthogonal experiment and analysis of component as well as properties. The polysaccharlde was extracted with hot water, depigmentation with acticarbon, precipitated with ethanol. The optimum extracting condition was ascertained by perpendicular experiments. The protein in polysaccharide was removed by Sevage' s way. The polysaccharide was purified by DEAE-32 and Sephadex G-25 chromatography. The purity of which was identified by the cellulose acetate membrane electrophoresis,its components analysed by paper chromatography. The optimum isolating condition was at 80 ℃ ,in 10 times water and for 3 h extraction. It was proved that the polysaccharide I of Potamogeton malaianus was composed of D-fructose and D-xylose, but the polysaccharide 11 included D-glucose and D-fructose except for an unknown constituent. This experiment could provide theoretical foundation for future investigation and utilization of Potamogeton malaianus.
