摘要
采用荧光光谱研究了一支箭凝集素(OPA)在不同温度,pH值以及不同淬灭剂作用时,其活性和分子构象变化的关系,结果表明:OPA具有较强的温度和酸碱度的耐受性,分子构象也无较大变化,但在pH 12时,其分子构象发生较大变化,活性也基本丧失.荧光淬灭剂丙烯酰胺和琥珀酰亚胺对OPA中Trp淬灭程度达100%,KI淬灭约62.5%的Trp荧光.表明OPA发射荧光的氨基酸残基大部分位于分子表面,少部分位于分子内部疏水环境中,且发荧光氨基酸所处微环境带电荷.
Fluorescence spectra was used to elucidate the relationship between the activity and molecular conformation of Ophioglossum Pedunculosum agglutinin (OPA) under different temperature , pH Value and quencher. The results showed that OPA was stable up to 60 ℃ and exhibit maximum activity between pH 4 and 8. Fluorescence quenching study of OPA by acrylamide, KI and Succinimide suggested that they can quench 100 %, 62.5 % and 100 % of the intrinsic fluorescence of tryptophan in OPA respectively, which indicated that acrylamide is the most efficient quencher and nearly all the tryptophan residues of OPA were located on the surface of the molecule.
