摘要
目的:探讨针刺头部腧穴对脑出血大鼠白介素-1β(IL-1β)表达的影响。方法:选取健康雄性Wistar大鼠160只,先随机分为3个组:模型对照组(简称模型组),模型十针刺组(简称针刺组),模型十脑复康组(简称脑复康组),每组50只大鼠,每组又分为术后6小时、1天、3天、7天5个时相点,每个时相点10只大鼠,制备脑出血模型,另设10只正常大鼠作为空白对照组(简称正常组)。在光镜下观察各组大鼠脑组织形态学改变。运用免疫组化及原位杂交的检测方法,观察不同时相点针刺对脑出血大鼠IL-1β的表达。结果:针刺组及模型组大鼠IL-1β阳性细胞数及阳性细胞平均光密度值随不同时相点的变化而逐渐增加,至3天时达高峰,7天时有所下降。在相同时相点,针刺组与模型组比较,IL-1β表达均明显低于模型组,两者比较差异有统计学意义(p<0.01)。结论:针刺头部腧穴能够抑制脑出血大鼠血肿周围脑组织IL-1β的表达。
Objective: To research the influence of the expression of IL-1β on the intercerebral hemorrage rat by acupuncturing scalp cave. Methods:The 160 healthy Wistar maid rats were selected. The rats were divided into 3 groups randomly:model group,pinprick group,piracetam group, 50 rats in each group. Each group was again divided into five time phase point of 6 hours, 1dy, 2days, 3days, 7days, 10 rats in each group. All rats were made to intercerebral hemorrage model. Another 10 rats were assigned as the matched blank group. We observated the change of brain histomophology of each group in deferent time phase point with light microscope. Observated the expression of IL-1β of the intercerebral hemorrage rat with the methods of immunity histochemistry and in situ hyybridizatiorL Results: In the model group and the pinprick group, the expression of IL-1β in rat's brain tissue surrounding the hematoma was increased gradually and to the peak in 3 days with the increasing of different time phase point, but decreased gradually in 7days. In the same time phase point, compare to the model group, the pinprick group expression of IL-1β was lower obviously. Both were relatively showed the significant difference (p〈0.01). Conclusions: Acupancturing scalp cave on the rat of the experiment intercerebral hemorrage inhibit the expression of IL-1β in rat's brain tissue surrounding the hematoma.
