小檗碱对2型糖尿病大鼠肺泡和腹腔巨噬细胞氧化低密度脂蛋白及脂蛋白脂酶的影响

摘要 目的探讨小檗碱对2型糖尿病大鼠血脂及肺泡和腹腔巨噬细胞氧化低密度脂蛋白(oxidized low-density lipoprotein,ox-LDL)、脂蛋白脂酶(lipoprotein lipase,LPL)表达的影响。方法SD大鼠分4组,正常对照组、高脂组、糖尿病组、小檗碱治疗组,治疗结束后测定各组大鼠血清葡萄糖、胰岛素和血脂含量,并检测肺泡巨噬细胞和腹腔巨噬细胞ox-LDL含量和LPL mRNA的表达水平。结果(1)小檗碱对糖尿病大鼠具有降低血糖、胰岛素水平和调节脂代谢紊乱等作用;(2)经小檗碱治疗后糖尿病大鼠肺泡和腹腔巨噬细胞ox-LDL含量下降,LPL mRNA表达降低。结论推测小檗碱对糖尿病大鼠的治疗作用可能与提高胰岛素敏感性,降低巨噬细胞ox-LDL、LPL表达有关。 Objective To study the effect of berberine on the expression of oxLDL,LPL in alveolar and peritoneal macrophages in type 2 diabetic rats. Methods Sprague-Dauley rats were divided into four groups: normal control group, high fat diet group, type 2 diabetes group and berberine treatment group. After treatment, serum glucose, insulin and lipid concentration were measured in different rats'groups. Then the level of oxLDL and LPL mRNA expression were detected in the alveolar macrophage (AM) and peritoneal macrophage (PM). Results The levels of serum glucose, insulin and lipid in type2 diabetic rats were decreased by berberine treatment. In the treatment groups, the level of oxLDL and LPL mRNA expression were reduced in AM and PM. Conclusion Berberine can enhance the sensibility of serum insulin and reduce the level of LPL expression in macrophage, which may be beneficial to diabetes.
