摘要
用碱水解后再用乙醇分级分离,从地龙中获得抗凝血酶活性。该活性不能被AT—Ⅲ抗体及鱼精蛋白中和,表明其活性表达不依赖于AT—Ⅲ。活性提取液在216nm处有单一吸收峰,费林试验阴性,双缩脲试验阳性,提示该活性不属多糖或蛋白质,而可能是一种耐碱耐热的小肽或含双键化合物,与肝素及其类似物、水蛭素等抗凝物质不相同。
By means of a procedure of alkaline hydrolysis followed by alcohol fractiona - tion, an antithrombin activity was obtained from earthworm. Both anti-AT-Ⅲ antibodies which almost completely abolished the antithrombin effect of heparin and protami n could not quench the abstracted antithrombin activity, suggesting in the nature of AT-Ⅲ independent. The solution containing the antithrombin activity showed a single absorption peak at 216nm, a positive biuret reaction and, after acid hydrolysis, a negative reaction with Fehling's reagent. These results indicate that the abstracted antithrombin actvity is neither polysaccharide nor protein and is different from the other anticoagulants known so far.
