摘要
研究了不同摇瓶条件对米根霉发酵体系中乳酸脱氢酶(LDH)与乙醇脱氢酶(ADH)的酶活变化,以及乳酸和乙醇转化率的影响,结果表明:当装液量从10%增加到30%,乳酸的质量浓度及相应的LDH活力呈下降趋势,而乙醇质量浓度及ADH活力随着装液量的增加而提高。摇床转速为220r/min时,乳酸质量浓度最大,对应的LDH活力也最大;摇床转速为160r/min时,乙醇质量浓度与ADH活力较高。单膜封口发酵乳酸质量浓度与LDH活力分别比双膜封口发酵提高64.4%和49.9%,而双膜封口发酵时,乙醇最大质量浓度与最大ADH活力分别比单膜封口发酵提高30.0%和25.5%。
The effect of different aeration conditions on fermentation product composition and correlative enzymes activities were investigated, the results indicated that the lactate production and lactate dehydrogenase (LDH) activity were both increased as the medium volumes increased from 10 % to 30 %, hut the alcohol concentration and alcohol dehydrogenase (ADH) activity were both decreased. The maximum production of lactic acid and LDH activity was achieved in cultures shaken at 220 r/min while 160 r/min shaking speed resulted in a higher alcohol concentration and ADH activity. The lactic acid concentration and the activity of LDH were 67.5 g/L and 5.83 U/mL, respectively, which increased by 64.4 % and 49.9 % respectively by single film fermentation. The ethanol concentration and the activity of ADH increased by 30.0 % and 25.5 % respectively, by dual film sealed fermentation compared with single film fermentation.
