染料木素对去卵巢大鼠基底前脑神经元型一氧化氮合酶神经元影响的体内研究

Study on the Effect of Genistein on Basal Forebrain Neuronal Nitric Oxide Synthase Neurons of Ovariectomized Rats in vivo

摘要 目的研究染料木素对去卵巢大鼠基底前脑神经元型一氧化氮合酶(nNOS)神经元的影响。方法对雌性大鼠双侧卵巢切除2周后行Genistein和雌激素替代治疗4周。用免疫荧光染色、RT-PCR和Western blot方法对nNOS神经元数量、nNOS基因和蛋白的表达量进行检测。结果去卵巢6周后,基底前脑内侧隔核(MS)和斜角带核垂直支(VDB)部位的nNOS神经元数量、nNOS基因和蛋白的表达量均明显减少,差异有统计学意义(P<0.05);雌激素和Genistein替代治疗后能显著增加去卵巢大鼠该部位的nNOS神经元数量、nNOS基因和蛋白的表达量,差异有统计这意义(P<0.05)。结论Genistein对去卵巢大鼠基底前脑nNOS神经元具有类似雌激素样神经保护作用。 Objective To study the effects of Genistein on basal forebrain nNOS neurons of ovariectomized rats. Methods Young female rats were ovariectomized bilaterally and two weeks later, performed Genistein and estrogen replacement therapy for four week. The nNOS neurons were detected by immunofluorescence staining. The nNOS mRNA and protein were detected by RT-PCR and Western blot respectively. Results Six weeks later, the number of nNOS neurons, the expression of nNOS mRNA and protein were significantly reduced in the medial septal nucleus (MS) and vertical limb of the diagonal band of Broca (VDB) in OVX rats. Estrogen and Genistein replacement therapy could prevent a decrease in the number of nNOS neurons and the expression of nNOS mRNA and protein. Conclusion The findings indicate that Genistein has neuroprotective effect similar to estrogen on basal forebrain nNOS neurons.
