棉铃虫对转Bt棉抗性遗传的AFLP连锁图谱分析 被引量:1

摘要 利用AFLP分子标记方法,对抗Bt棉棉铃虫(Helicoverpa armigera)抗性品系和敏感品系回交BC1代(F1(YCR♀×YCS♂)♀×YCR♂)群体进行连锁图谱的构建。经23组AFLP引物的选择性扩增,共得到406个多态位点,√检验后有217个为有效位点。利用Mapmaker/Exp(Version3.0b)软件进行连锁分析,其中126个标记分属35个连锁群,连锁群标记数变化范围为2~11个,平均每个连锁群标记数为3.6个,该图覆盖的基因组长度为1138.4cM(图距单位),连锁群长度变化范围为4.0~166.1cM,连锁群的平均长度为32.53cM。连锁图谱的构建为棉铃虫对抗Bt棉抗性基因的定位奠定了基础。 AFLP makers were mapped using BC1 ( F1 ( YCR♀×YCS♂)♀×YCR♂ ) population of Helicoverpa armigera (Htibner). In this study 23 AFLP primer combinations were used and 406 polymorphic markers were detected, with 217 valid loci consistent with the expected segregation ratio of 1:1 at 0. 05 significant level. 35 linkage groups with 126 loci were mapped by using Mapmaker/Exp (Version 3.0b). The number of loci mapped in the 35 groups was at a range of 2 11 and there were 3.6 (average value) loci per group. The total length of the groups was 1 138. 4 cM. The lengths of the 35 groups varied at a range of 4. 0 - 166. 1 cM and the average length of groups was 32.53 cM. A linkage map of Helicoverpa armigera has laid the foundation for positioning the gene of resistance to Bt.
