姜黄素、槲皮素、尼莫地平对C6胶质瘤细胞替尼泊苷敏感性的影响* 被引量:1

摘要 目的:观察姜黄素(Curcumin,Cur)、槲皮素(Quercetin,Que)和尼莫地平(Nimodipine,Nim)对胶质瘤细胞化疗药物替尼泊苷(Teniposide,Ten)敏感性的影响。方法:体外培养大鼠脑胶质瘤C6细胞,采用MTT比色法确定Ten的IC 50及Cur、Que、Nim的安全浓度,检测Ten与Cur、Que、Nim联合使用对肿瘤细胞增殖率的影响。结果:Ten的IC50为0.5μg/ml,Cur、Que和Nim的安全浓度分别为5.16μg/ml、0.03μg/ml和10μg/ml。Ten分别与Cur、Nim联合使用对胶质瘤细胞的抑制率明显高于单独使用Ten的抑瘤效果,Ten与Que联用的协同作用最明显。结论:Cur、Que和Nim能增强大鼠脑胶质瘤C6细胞对化疗药物Ten的敏感性,合用呈现协同效应。 Objective: To investigate the effects of curcumin (Cur), quereetin(Que) and nimodipine(Nim) on the chemotherapy sensitivity of glioma C6 cells to tenipeside and to provide some useful information for chnic chemotherapy. Methods: C6 cells were cultured in DMEM containing 10% fetal bovine serum. MTT colorimetry assay was performed to determine the median inhibitory concentration (IC50) of Ten and the safe concentrations of Cur, Que and Nim. Then the inhibition effects of Ten in combination with Cur, Que, Nim on the C6 cells were evaluated, respectively. Results: The IC50 of Ten is 0.5 μg/ml and the safe concentrations of Cur, Que and Nim are 5.16 μg/ml, 0.03 μg/ml and 10 μg/ml, respectively. The inhibition effect of Ten in combination with Cur or Nim is evidently better than that of Ten alone, especially with Que. Conclusion: Cur, Que and Nim could enhance the chemotherapy sensitivity of glioma C6 cells to Ten, and they showed synergistic interactions.
