摘要
目的利用人转录因子siRNA文库筛选凋亡相关siRNA,为发现新的凋亡相关基因奠定基础。方法人转录因子siRNA文库稳定转染Jurkat细胞后用rhTRAIL诱导,使用高通量siRNA文库筛选法筛选出具有抗凋亡特性的细胞群。检出细胞群中的siRNA表达片段,逐个验证其抗凋亡功能。结果筛选出了具有抗凋亡特性的细胞群。克隆并鉴定了部分具有抗凋亡作用的siRNA表达片段。结论利用人转录因子siRNA文库进行了有效地高通量筛选,筛选出了具有抗凋亡能力的siRNA表达片段。
Objective Screening of human transcription factor siRNA library for apoptosis related siRNAs, to facilitate the further identification of novel apoptosis related genes. Methods Jurkat cells were stably transfeeted with human transcription factor sIRNA library. After being treated with rhTRAIL, cells with anti-apoptosis capacity were selected by high-throughput siRNA library screening method, siRNA expressing cassettes were recovered from the cells and verified individually for their anti-apoptosis ability. Result Cells with anti-apoptosis capacity were enriched, siRNA expressing cassettes were cloned and some of them were verified to be able to anti-apoptosis. Conclusion High-throughput screening was done with human transcription factor siRNA library. And some siRNA expressing cassettes with anti-apoptosis capacity were identified.
