摘要
目的观察替普瑞酮(geranylgeranylacetone,GGA)对持续热损伤大鼠、中暑大鼠的保护作用。方法大鼠测定肛温(直肠温度,core temperature,Tco)后持续暴露于41.0~41.5℃热仓中建立持续热暴露模型或暴露于41.0~41.5℃热仓中35min建立中暑模型,观察连续3d灌胃给予GGA(100mg/kg)后大鼠直肠温度变化及在热仓中的存活时间或热损伤后在室温中的存活时间。结果热暴露组大鼠进入热仓后体温上升迅速,肛温到达42.1℃的时间约为24min,在热仓中的生存时间约为46min;GGA(100mg/kg)连续3d给药对大鼠基础体温无影响,但能轻微延长大鼠进入热仓后体温到达42.1℃的时间,约为26min(P<0.05),而对大鼠体温从42.1℃到死亡的时间及在热仓中的总生存时间均无明显影响(P>0.05)。正常大鼠热损伤35min后在室温环境中的存活时间约为20min,而GGA预处理后在室温环境中的存活时间延长至195min(P<0.01)。结论GGA对持续热暴露条件下大鼠热损伤无保护作用,但明显延长大鼠中暑后在室温环境中的存活时间。
Objective To evaluate the protective effects of geranylgeranylacetone (GGA) on hyperthermia- treated rats. Method Two different models of hyperthermia were used. In the continuous hyperthermia model, rats were continuously exposed to 41.0-41.5℃ and the survival times were determined. In the heat stroke model, rats were exposed to 41.0-41.5℃ for 35 minutes to induce heat stroke. Effect of GGA was determined by treating the rats with an oral dose of GGA (100 mg/kg/d for 3 consecutive days) 3 days before the start of heat treatment. After treatment, the rats were allowed to recover at room temperature (24℃). Tco and the survival time were determined. Result Under the condition of continuous hyperthermia, Tco was rapidly increased. It took about 24 minutes to reach 42.1℃ and the survival time was 46 min. In the group of rats with GGA treatment, the time for Tco to reach 42.1℃ was slightly prolonged to 26 min (P〈0.05). GGA pretreatment had no effect on the basal temperature and the survival time (P〉0.05). In the heat stroke model (35 minutes of heat treatment), the survival time of normal rat was :20 minutes. GGA was found to prolong the survival time of heat stroke rats to 195 minutes (P〈0.01). Conclusion GGA has no protective effect on rats under the conditions of continuous hyperthermia. GGA can prolong the survival time of rats under the heatstroke condition.
