同种内耳抗原免疫孕豚鼠所产子鼠听觉损伤程度与母鼠特异性免疫反应水平相关性研究 被引量:5

Analyses of the relationship between the level of special immune reaction of the females immunized with inner ear antigens and the degree of their offspring's hearing loss

摘要 目的:探讨自身免疫性感音神经性聋孕豚鼠所产子鼠听觉损伤(听损)程度与母鼠特异性免疫反应水平的相关性,同时了解子鼠内耳病理损伤和听损的特点。方法:同种粗制内耳抗原免疫雌性豚鼠,后将其与雄鼠交配,妊娠并生产子鼠。测试母鼠特异性抗体、细胞免疫反应及听觉功能,光镜观察内耳组织形态变化;测试全部出生后2周和部分出生后6周子鼠听觉功能,并亦用光镜观察内耳组织形态变化。结果:母鼠抗体水平在妊娠中期达峰值,特异性细胞免疫反应水平在妊娠晚期尚在进一步升高。子鼠听神经复合动作电位(cAP)阈值与母鼠血清特异性抗体水平及cAP阈值升高程度呈线性正相关、与母鼠特异性细胞免疫反应水平呈非线性2次多项式关系。结论:母鼠血清特异性抗体水平和cAP阈值升高程度越显著,子鼠发生听损概率越高,听损亦越严重。随出生后时间延长,子鼠听损无明显变化,听损子鼠可发生膜迷路积水。 To explore the relationship between the level of special immune reaction in female guineapigs immunized with conspecific crude inner ear antigens (CIEAgs) and the degree of its offspring's hearing loss,and at the same time, to explore the characteristics of inner ear histopathology and hearing dysfunction in the offsprings, nethotis The female guinea-pigs were immunized by CIEAgs, and then copulated with male guinea-pigs and produced offspring. Before and after immunity, the female guinea-pig's special antibodies against inner ear antigens in serum were measured in series using ELISA method and the level of special cellular immune reaction by 3 H-TdR-lymphocyte transform test. The hearing function of all female guinea-pigs and offspring were tested with electrocochleography. In the end, all animal's temporal bones histological slices were observed under light microscope. Results After immunized with inner ear antigens, level of humoral special antibodies in serum and special cellular immune reaction rose in some female guinea-pigs. The and cellular imunizty peaked at the middle and last stage of pregnancy. A positive-relativity between the threshold of offspring guinea-pigs and their mother's special antibodies level in serum or acoustic nerve compound active potential(cAP) threshold were found. There were nonlinear twice polynomial relationship between the threshold of off- spring guinea-pigs and their mother's special cellular immune reaction. Conclusion The higher the female guinea-pig's antibodies level in serum and cAP threshold, the more serious and more often their offspring's hearing loss. Along with postnatal time prolonging,the offspring guinea-pig's hearing dysfunction do not evidently change. The endolymphatic hydrops happens in offspring guinea-pigs with heating loss.
