肝素对大鼠胰岛细胞保护作用的实验研究 被引量:1

摘要 目的:探讨胰腺切取前全身肝素化对大鼠胰岛细胞的保护作用。方法:Wister大鼠20只被随机分为两组,肝素化组胰腺切取前经门静脉注入肝素3 mg/kg,全身肝素化后切取胰腺,非肝素化组胰腺切取前经门静脉注入等容量生理盐水,采用酶消化法分离胰岛,双硫腙(DTZ)染色鉴定并记数胰岛细胞,AO-PI染色计算存活率,胰岛素释放试验检测胰岛细胞功能。结果:肝素化组:胰岛细胞的产量为每克(897±169)个,纯度86%,存活率90%,胰岛素释放试验高糖组为(506±96.36)μIN/mL。低糖组为(329±59.17)μIN/mL;非肝素化组:胰岛细胞的产量为每克(620±150)个,纯度70%、存活率85%,胰岛素释放试验高糖组为(300±50.23)μIN/mL低糖组为(200±45.12)μIN/mL。两组对应指标比较P<0.05。结论:肝素对胰岛细胞具有保护作用,胰腺切取前全身肝素化可以作为预处理手段应用于胰岛细胞移植。 Objective:To invesigate the action of heparin on rat islet cell. Methods : Wister rats were randomized into two groups:heparin group and the group of non-heparin,heparin group. In heparin group, heparin was injected at the dose of 3 mg/kg by the way of portal vein before pancreatic resection. In no-heparin group ,heparin was replaced by saline. Methods:Of enzyme digestion discontinuous gradient centrifugation were employed to isolate and purify islet cells. DTZ staining and AO-PI staining were performed to differentiate valid islet cells. Insulin release tests were carried out to examined the function of islet cell. Results:In heparin group ,islet cells production of (897±169)/g,purity:86%,survival rate 90%. Insulin release test:the low glucose group (329±59.17) μN/mL;high glucose groups: (506± 96. 36) μIU/mL. Non-heparin group islet cells production of (620±150)/g,purity 70% ,survival rate 85% insulin release test high glucose group (300±50.23) μIN/mL the low glucose group (200±45.12)μIN/mL. Corresponding indexes of two groups were compared,P〈0. 05. Conclusion :Heparin possess significant protective effect on islet cells ,pre-resection systemic heparin can be used as an effective pretreatment method in the islet cell transplantation
