摘要
目的探讨大鼠海马区及皮层EphA4表达在慢性脑缺血的不同时间点的动态变化。方法大鼠随机分为正常组、假手术组、15d、30d和60d模型组.采用双侧颈总动脉永久结扎建立慢性脑缺血模型;Morris水迷宫试验检测行为学变化;免疫组化染色检测EphA4的表达变化。结果EphA4免疫反应阳性细胞数模型组比假手术组均增多(P<0.05),与行为学改善基本相符。结论慢性脑缺血造成EphA4表达持续升高可能是成年哺乳动物损伤后神经再生障碍的重要原因之一。
[Objectives] To explore the dynamic changes of EphA4 expression in the cortex and hippoeampus after chronic cerebral ischemia in rats. [Methods] The rats were divided into five groups randomly: normal group, sham operation group, 15 day, 30 day and 60 day model groups. The chronic cerebral isehemie models were produced by the permanent occlusion of bilateral common carotid arteries. All of the rats were trained in Morris Water Maze to find the changes of etiology. The expression of EphA4 was measured by immunohistoehemistry method. [Results] The expression of EphA4 in all the model groups was significantly increased than in the sham operation group (P 〈0.05), which was in accordance with the behavioral observations. [Conclusion] The increased expression of EphA4 in the chronic cerebral isehemie lesions suggests that it may play an important inhibitory role in the neural regeneration.
