摘要
目的探讨基质金属蛋白酶2、9(MMP-2、MMP-9)在鼻咽癌组织中的表达及其意义。方法应用免疫组化S-P法检测MMP-2、MMP-9在正常对照组(7例)和鼻咽癌(45例)组织中的表达,45例鼻咽癌患者中发生颈部淋巴结转移有24例,其中Ⅰ～Ⅱ期21例,Ⅲ～Ⅳ期24例。结果鼻咽癌组织中MMP-2和MMP-9的阳性率明显高于正常对照组(P<0.05),其中淋巴结转移组阳性率比无淋巴结转移组表达明显增高(P<0.05)。在鼻咽癌标本中还发现癌巢周边的成纤维细胞、癌周浸润的单核细胞、淋巴细胞以及血管内皮细胞中也有MMP-2和MMP-9的表达。结论MMP-2、MMP-9可能在鼻咽癌发生发展和突破基底膜向外扩散及淋巴结转移中起到重要作用。
Objective To investigate the expression of matrix metalloproteinase--2（MMP--2） and matrix metalloproteinase-- 9 （MMP-- 9） in the nasopharyngeal carcinoma （NPC）. Methods The ex- pression of MMP--2 and MMP--9 was detected by immunohistochemical S--P technique in 45 NPC ca- ses and 7 non--cancer cases, which have been treated as control group. There were 24 cases with the metastasis of neck lymphoid nodes among the 45 NPC cases, including 21 cases in clinical stage Ⅰ-Ⅱ and 24 cases in clinical stage Ⅲ-Ⅳ. Results The expression rates of MMP--2 and MMP--9 in NPCs were obviously higher than those in control specimens （P〈0.05）. The expression rates of the cases with the metastasis of neck lymphoid nodes were obviously higher than those without metastasis. And the Immunostaining for MMP--2 and MMP--9 expression was seen in fibroblast, macrophages, lym- phocytes and vascular endothelial cells beside tumor cells. Conclusions MMP--2 and MMP--9 play an important role in the invasion and subsequent metastasis of NPC by degrading basement membrane components.
