摘要
目的:研究中药生大黄、丹参对实验性重症急性胰腺炎(ESAP)病程早期小肠天然免疫屏障的保护作用。方法:取100份实验条件下存活的SD大鼠样本,每组10份,分为假手术组(SO),改良Aho法造模的模型组(SAP12、SAP24、SAP36),大黄治疗组(DH12、DH24、DH36),大黄丹参治疗组(DD12、DD24、DD36),用Quanty One软件分析PCR产物凝胶图像以检测各组Dn-5 mRNA的表达变化。结果:模型组各时段与正常大鼠的回肠末端Dn-5 mRNA相比,表达明显下调并随时间延续加剧;而各中药干预组与模型组比较回肠末端Dn-5mRNA有统计学意义的上调。结论:中药生大黄和丹参在SAP发病过程中增强肠黏膜天然免疫屏障作用。
Objective To examine the changes of intestinal defensin-5 mRNA expression in SD rats with experimental severe acute pancreatitis(ESAP) and explore the part of mechanisms of bacterial translocation at the molecular level, and to investigate the effect of activating blood circulation and dispelling stasis herbs(Rhubarb, Danshen Root) in model rats with severe acute pancrcatitis. Methods One htmdred samplcs of SD rats were divided into 10 experimental groups(SO, SAP12,SAP24, SAP36, DH12, DH24, DH36, DD12, DD24, DD36) with 10 samples each. ESAP was induced by modified Aho's method, thc rats were killed at 12, 24 and 36 hours after the injection. Thc control group(SO) was killed directly. The amplification products of RT-PCR were electrophoresed and the images were analyzed by Quanty One software. Results Compared with the SO group, each period of intestinal defensin-5 mRNA expression in model group was distinctively regulated-down and aggravated with ESAP-proceeding; while the one in Chinese herbal group was regulated - up with statistically significance. Conclusion The traditional Chinese herbs Rhubarb and Danshen root can restrain the bacterial translocation of intestine in SAP.
