摘要
目的建立柱前衍生化高效液相色谱法同时测定人唾液中腐胺和精胺含量的方法。方法唾液经纯化、丹酰氯衍生化后用苯提取,以Hypersil BDS CN为正相色谱柱,丙酮-氯仿(2:98)为流动相,流速1mL/min进行测定,检测波长为425nm。结果腐胺和精胺均在50~1000nmol/mL范围内线性关系良好,r分别为0、9991和0、9992,此法回收率分别为94.65%和90.48%,RSD分别为1.6%和2.1%。结论此法灵敏、稳定,重现性好,可检测人唾液中多胺含量。
Purpose To develop a high-performance liquid chromatographic method for the determination of polyamine in human saliva. Methods The derivation with dansyl chloride was followed by normal-phase chromatography with a mobile phase of acetone/chloroform (2:98)at a flow rate of 1 mL/min and with the detection wavelength set at 425nm. Results The standard curve was linear over the concentration range of 50-1 000 nmol/mL for putrescine and spermine with correlation coefficient 0. 999 1 and 0. 999 2. The recoveries were 94.65% with RSD 1.6% and 90.48% with RSD 2.1% for putrescine and spermine, respectively. Conclusion The present study provided a sensitive, accurate and reproducible method for the determination of polyamine levels in human saliva.
