尼罗罗非鱼Dmrt1基因克隆及在不同组织中的表达 被引量:5

Cloning of Dmrt1 Gene and Its Expression in Different Tissues of Oreochromis niloticus

摘要 Dmrt1基因是Dmrt基因家族的一个重要成员,在动物性别决定过程中发挥重要作用。参照小鼠Dmrt1基因DM保守区及有关文献,设计了1对简并引物,扩增了尼罗罗非鱼的Dmrt1基因,并对扩增产物进行了亚克隆与测序。结果在雌雄尼罗罗非鱼个体中各获得1个预期的基因片段,长度为140bp,序列无性别差异。序列同源性分析表明,尼罗罗非鱼Dmrtl基因DM盒区核苷酸序列与人和鼠相应基因的同源性为78%;编码的氨基酸序列与人和鼠相应基因编码序列的同源性为89%,充分显示了Dmrt1基因在进化上的高度保守性。进一步根据获得的Dmrt1序列,设计1对特异引物,采用RT-PCR技术对尼罗罗非鱼不同组织Dmrt1基因的表达进行了研究。结果发现,Dmrt1只在精巢中特异表达,在卵巢、眼、肠、鳃及心脏组织中无表达,提示该基因在性别分化和功能维持上可能具有重要作用。 The Dmrt1 gene plays important roles in sex determination. The 140bp DM -box of Dmrt1 gene in Oreochromis niloticus was amplified by using degenerated PCR,and then subcloned and sequenced.The sequences from male and female O. niloticus were compeletely identical.Compared with the human and mouse Dmrt1 gene,the sequences of Dmrt1 shared 77%nucleotide homology and 89% amino acid indentity. The results indicated that Dmrt1 genes were highly conservative in phylogeny. According the sequence of Dmrt1, specific primers were designed for RT-PCR in different tissues in adult O. niloticus. Dmrtl expression was only detectable in testis, and no expression was detected by RT - PCR in the following tissues: ovary, eye, intestine, gill and heart.
