摘要
对粗柄羊肚菌(Morchella crassipes)原生质体形成、再生条件进行了研究。原生质体形成的合适条件:酶解系统为4%蜗牛酶、5%溶壁酶、4%纤维素酶;缓冲系统为0.6mol/LKCl的磷酸缓冲液(pH6.98),酶解时间4h,酶解温度30℃,菌龄56h;原生质体产量为5.3×104个/mg。经0.3mol/LKCl+0.3mol/L环己六醇的PSA再生培养基培养,再生率为9%。
The protoplast formation and regeneration of Morchella crassipes were studied. The optimum conditions for protoplast formation were determined. In details, the enzymolysis system consisted of 4% snailase, 5% lywallzyme and 4% ellulase. And 0.6 mol/L KCl phosphate buffer (pH 6.98) was used. The enzymolysis was conducted at 30℃ for 4 h. The age of cells for protoplast preparation was 56 h. The protoplast yield reached 5.3×10^4 cells/mg. On the PSA (potato sucrose agar) medium contenting 0.3 mol/L KCl and 0.3 mol/L inositol, the regeneration rate was 9%.
