摘要
目的:利用高效毛细管电泳(High Performance Capillary Electrophoresis,HPCE)观察低硒对发育期大鼠脑核基质蛋白(Nuclear Matrix Proteins,NMPs)的影响。方法:利用人工配制的低硒饲料喂养的SD大鼠,分为对照组和低硒组。取第三代出生后21天的大鼠,处死后迅速取脑,分离大脑皮层。提取NMPs,经初步鉴定后,用HPCE的毛细管区带电泳(CZE)模式进行NMPs的分离,观察其组成成分的改变。结果:NMPs经HPCE分离出3~4个蛋白峰,波形较稳定,在进样9min左右有一个蛋白峰低硒组明显低于对照组。结论:经HPCE分析能够观察到低硒脑NMPs的变化。
Objective:To study the alteration of nuclear matrix proteins(NMPs) of developing low-selenium rat brain by High Performance Capillary Electrophoresis(HPCE).Methods:Low-selenium group and control group of SD rats were feed with Low-selenium and selenium adequate food respectively and generated.Rats of the third generation at 21 days of age were killed and the cortexes were dissected on ice rapidly.NMPs of both group was extracted and analyzed by HPCE.Results:The NMPs can be separated by HPCE with three or four peaks.There was one peak,which occurred at about 9 min separation,showed significant drop in the low-selenium group.Conclusion:The alteration of NMPs of low-seleniumcy rat brain can be induced by HPCE.
