摘要
目的:以陕西洛南产北细辛为研究对象,采用反相高效液相色谱法测定细辛挥发油中甲基丁香酚及黄樟醚含量,以满足质量控制的要求.方法:以甲醇-水(80∶20)为流动相,流速1mL/min,经Inertsil ODS-3(4.6mm×250 mm,5μm)色谱柱分离,于250nm波长检测.结果:甲基丁香酚及黄樟醚的分离效果良好.甲基丁香酚在19.28~462.72mg/L浓度范围内呈良好的线性关系,回归方程为:Y=46156.59X+32840.02(r=0.9999,n=5);黄樟醚在11.78~471.20mg/L浓度范围内呈良好的线性关系,回归方程为:Y=29591.52X-75017.20(r=0.9999,n=5).甲基丁香酚和黄樟醚的平均回收率分别为99.95%和100.03%,相对标准偏差分别为0.97%和0.68%.结论:本法可同时测定细辛挥发油中甲基丁香酚及黄樟醚的含量,为质量控制提供依据,方法简便、快速,准确.
AIM: To establish a reversed-phase high performance liquid chromatographic (RP-HPLC) method for simultaneous determination of methyleugenol and safrole in the essential oil of Asarum. METHODS: The mobile phase was methanol-water (80:20) , with the flow rate as about 1 mL/min. Inertsil ODS- 3 (4.6 mm ×250 mm, 5 μm) column was used and the detection wavelength was 250 nm. RESULTS: The calibration curves was linear in the concentration range of 19.28-462.72 μg/mL for methyleugenol. The regression equation: Y = 46156.59X+32840.02 (r=0.9999, n=5). The calibration curves was linear in the concentration range of 11. 75- 471.20 mg/mL for safrole. The regression equation: Y = 29591.52 X-75017.20 (r=0.9999, n=5). The average recovery of methyleugenol and safrole was 99.95% and 100.03% respectively. The relative standard deviations (RSD) were 0.97% and 0.68% respectively. CONCLUSION: This method is simple, quick and accurate for the simultaneous separation and determination of methyleugenol and safrole.
