摘要
目的:研发一种布鲁氏菌病快速诊断试剂。方法:应用胶体金免疫层析法(GICA),经布鲁氏菌病病人和健康人不同血清用量的测试,选择最佳血清用量,建立快速检测诊断布鲁氏菌病的新方法。结果:胶体金免疫层析法快速检测布鲁氏菌病最佳血清用量为10μl,检测布鲁氏菌病敏感性和特异性分别达到93.6%和94.9%,youden指数为0.885,检测结果与DIGFA及SAT比较三者无统计学意义(P>0.05)。结论:应用胶体金免疫层析法(GICA)快速诊断布鲁氏菌病,不仅具有试剂敏感、特异的优点,且血清用量更少(10μl),操作更为简便,更适合于布鲁氏菌病的诊断、流行病学调查及现场应用。
Objective :To establish a new method for rapid diagnosis for human brucellosis. Methods:The Colloidal Gold - Immunochromatographic Assay was developed through detecting on the positive serum of human brucellosis and healthy people respectively. The new method was based on the optimal condition which was selected after analyzing the different volume serum for detection. Results:10 μl is the optimal volume for detection. The sensitivity and specificity of Colloidal Gold - Immunochromatographic Assay for detecting human brucellosisis is 93.6% and 94. 9% respectively, the youden index was 0. 885, and there were no significant difference among Colloidal Gold - Immunochromatographic Assay, SAT and DIGFA (P 〉 0. 05 ). Conclusion:We established a rapid method for brucellosisis diagnosis, which is Gold - Immunochromatographic Assay, and it is not only sensitivity and specificity, but also less detected sample needed( 10 μl). This new method is a simple way, and more suitable for the diagnosis, epidemiology investigation and field application of brucellosisis.
