互花米草盐胁迫应答基因的相关序列扩增多态性分离研究

摘要 设计合成了12对相关序列扩增多态性(SRAP)引物,采用SRAP技术从互花米草根中克隆了受盐胁迫诱导的DNA片段,分析了基因表达的差异。结果表明,对照组和盐胁迫组的遗传多态性高达82%。同时,本文采用Northern杂交分析了互花米草中的一个差异表达片段RGG,RGG cDNA测序及GAPPED分析结果表明,RGG片段由564个碱基组成,其编码的氨基酸序列与水稻的α-2,4-葡糖化酶之间存在69%的相似性。研究结果表明,植物RGG基因诱导表达可能是植物对逆境胁迫的一种适应性反应。 Twelve pairs of sequence related amplified polymorphism primers(SRAP) were synthesized.In this paper,SRAP primers were used to identify genes differentially expressed in the roots of smooth cord-grass under salt stress.The results indicated that the percentage of polymorphism was about 82% between the transcript of the control and the salt treatment.RGG differentially expressed fragment was verified by Northern blot.Gene sequencing and GAPPED analysis showed that the fragment was 564 bp long and its encoded amino acid sequence had 69% identity with α-2,4-glucase.The results indicated that genes abduction expression of RGG might be a certain adaptive reaction on the adversity stress for the plants.
