血清饥饿法诱导人角质细胞HaCaT细胞周期同步化的研究 被引量:2

摘要 探讨血清饥饿法对人表皮细胞HaCaT细胞周期的影响.采用无血清和低血清浓度的DMEM培养基饥饿HaCaT细胞,分别培养6h,12h,22h时,用倒置显微镜观察细胞形态,并且收集各组细胞,用流式细胞仪分析细胞周期各个时相细胞百分比.结果发现HaCaT细胞系在含0.2%FBS的DMEM培养基中培养12h,细胞仍然贴壁生长,并且可以使G0/G1期细胞百分率达到66.5%.因此,血清饥饿法可以应用于人表皮细胞HaCaT同步于G0/G1期,从而为后期药物筛选打下基础. We have determined the effect of cell cycle in human normal keratinocyte line HaCaT by serum starvation in the study. By using serum-free and low concentration serum DMEM culture medium, cell line HaCaT was cultured; and the cells were collected in 6h ,12h ,22h, respectively, then cell form was observed by inverted microscope and the results were analysed by flow cytometry. After HaCaT cells in DMEM culture medium containing 0.2% FBS was cultured for 12 h, the cells were adherent growth and 66.5% at G0/G1 phase. The human keratinocytes line HaCaT may be synchro- nized in G0/G1 phase by serum starvation, so the basement was grounded for later medical filtration.
