摘要
为给体细胞核移植提供更多的G0/G1期供体细胞以提高克隆的成功率,本试验采用不同的接触抑制和血清饥饿时间及含羞草酸(m imosine)不同浓度和时间3种方法对麋鹿皮肤成纤维细胞进行同期化处理,然后利用流式细胞仪分析细胞周期。结果表明,接触抑制24 h G0/G1期比例可达91.95%;血清饥饿24 h有89.42%的细胞处于G0/G1期,当处理时间超过144 h细胞凋亡和异常聚集的比例会增加;适宜浓度(0.4 mmol.L-1)的含羞草酸处理24 h能使麋鹿成纤维细胞同步化至G0/G1期,但易引起细胞损伤。
In order to provide more G0/G1 stage cells for valuable or endangered Elaphursu davidianus inter-species cloning, the cell cycle of E. davidianus fibroblasts was synchronized by contact inhibition, serum starvation and mimosine. Cell cycle was analyzed by flow cytometry. The percentage of G0/G1 stage cells increased with the extent of confluence. The results showed that 91.95% of the fibroblast cells were in the G0/G1 stage after contact inhibition for 24 h. The percentage of G0/G1 stage cells was increased by serum starvation effectively. 89.42% fibroblast cells were synchronized to G0/G1 stage after 24 h treatment, but the percentage of apoptosis and aggregates increased after 144 h. Fibroblast cells were synchronizated by 0. 4 mmol ·L^-1 mimosine for 24 h, too. But mimosine could cause damage to fibroblast cells. The data may be important in choosing optimal method for cell cycle synchronization and improve efficiency of somatic cell nuclear transfer.
