摘要
目的研究室旁核注射apelin-13对胃缺血/再灌注(GI/R)损伤的作用及细胞机制。方法室旁核注射apelin-13,制备GI/R模型;用Western-blot方法检测胃黏膜细胞的凋亡以及凋亡相关基因c-Jun氨基末端激酶(c-Jun N-terminal kinase,JNK)的磷酸化水平。结果与单纯GI/R组相比,室旁核注射apelin-13能明显增加GI/R后胃黏膜细胞的损伤,同时可以升高caspase-3蛋白表达及JNK的磷酸化水平。结论室旁核注射apelin-13对大鼠GI/R损伤具有加重作用。
Objective To observe the effects and mechanism of apelin - 13 microinjection into the hypothalamic paraventricular nucleus (PVN) on gastric ischemia/reperfusion (GI/R) injury in rats. Methods Apelin - 13 was first microinjected into the PVN before the establishment of GI/R model, which was carried out in rats by clamping the celiac artery for 30 min and the following reperfusion for 1 h. Western - blot was used to determine the expressions of caspase - 3 and p - JNK of the gastric mucosal cells. Results Compared with the GI/R control group, apelin - 13 microinjection into the PVN markedly aggravated the damage in gastric mucosa and enhanced the expressions of caspase - 3 and p-JNK. Conclusion Apelin - 13 microinjection into PVN can aggravate the harmful effect on gastric mucosa induced by GI/R.
