电针对非酒精性脂肪肝肝细胞色素P450 2E1表达及氧化抗氧化的影响 被引量:13

摘要 目的:研究电针对非酒精性脂肪肝(NAFLD)肝细胞色素P450 2E1(CYP2E1)表达及氧化抗氧化的影响.方法:用高脂饲料诱导大鼠脂肪肝模型,同时给予电针治疗,观察肝组织病理形态的变化和肝CYP2E1表达.测定肝内丙二醛(MDA),超氧化物歧化酶(SOD),甘油三酯(TG),总胆固醇(TC)以及谷胱甘肽(GSH)含量的变化.结果:与脂肪肝模型组比较,电针组的肝组织脂肪变性和炎性损伤得以改善.免疫组化证明电针治疗能抑制脂肪肝肝细胞色素CYP 2E1的表达,同时肝内MDA,TG,TC显著降低(P<0.01),SOD,GSH活性升高(P<0.01).结论:电针通过抑制脂肪肝细胞色素CYP 2E1的表达,从而减轻脂质过氧化反应,增强抗氧化能力,使肝组织的脂肪变性和炎性损伤得以改善. AIM : To investigate the influence of electroacupuncture on cytochrome P450 2E1 ( CYP 2E1 ) expression, oxidation and anti-oxidation of non-alcoholic fatty liver tissue. METHODS: Fatty liver model of rat was induced with high fat diet. The pathological morphological changes and CYP2E1 expression in liver tissue were observed after electroacupuncture treatment. The content changes of intrahepatic malondialdehyde (MDA), superoxide dismutase (SOD), triglycerides (TG), total cholesterol (TC) and glutathione (GSH) were determined. RESULTS: Compared with fatty liver model group, fat denaturation and the inflammation in liver tissue were alleviated in electroacupuncture group. Immunohistochemical staining showed that electroacupuncture treatment inhibited the CYP 2E1 expression in fatty liver tissue. Liver MDA, TG, TC significantly reduced (P 〈 0.01 ). The activities of SOD and GSH increased (P 〈 0.01 ). CONCLUSION: Electroacupuncture can alleviate the lipid peroxidization through suppressing the CYP 2E1 expression in fatty liver tissue, so as to strengthen the anti-oxidization ability and alleviate the fat denaturation and the inflammation damage of liver tissue.
