摘要
目的:研究胡椒配伍甘草使用对大鼠肝细胞色素P450的影响。方法:灌胃给予SD大鼠甘草酸50mg/kg,胡椒碱30mg/kg,胡椒碱30mg/kg+甘草酸20mg/kg、50mg/kg、90mg/kg,苯巴比妥钠50mg/kg和等量生理盐水,7d后处死大鼠,差速离心法制备肝微粒体,采用Johannesen等[1]方法测定肝细胞色素P450含量。结果:与正常对照组(4.971±1.631nmol/ml)相比,胡椒碱组肝细胞色素P450含量(2.734±1.013mol/ml)明显降低,甘草酸组(4.540±1.094nmol/ml)无明显变化,苯巴比妥钠组肝细胞色素P450含量(16.620±3.503nmol/ml)明显升高,而胡椒碱+甘草酸组其肝细胞色素P450的含量(3.008±0.784、4.215±1.237、5.075±1.324nmol/ml)随甘草酸剂量的增加而增加。结论:胡椒碱可以明显抑制大鼠细胞色素P450的活性,甘草酸本身无明显诱导肝细胞色素P450的作用,但能对抗胡椒碱引起的肝药酶抑制,并具有剂量依赖性。
Object;To explore the influence on liver cytochromes P450 by co- administration of piperine and glycyrrhizic acid in rat. Method:56 SD rats were divided into 7 groups with 8 in each one. The rats were given glycyrrhizic acid 50 mg/kg i. g., piperine 30 rag/kg i. g., piperine 30 mg/kg + glycyrrhizic acid 20mg/kg、50mg/kg、90mg/kg i. g.;pheno-barital sodium 50mg/kg,respectively.The normal control group was given with 0.9% NaCl,i.g. .In day 7,all groups were abrosiaed for 24h. In day 8, all rats were killed. The microsome of liver was collected by centrifugaliztion and the concentration of cytochromes P450 was detected using Johannesen' s method. Result: Compare to normal control group(4.971±1.631nmol/ml), the concentration of cytochromes P450 was markedly decreased with piperine group(2.734±1.013nmol/ml). The concentration of cytochromes P450 of glycyrrhizic acid group(4.5040±1.094nmol/ml) had no difference. The group that co-administration with piperine and glycyrrhizic acid which concentration of cytochromes P450 was depened by the dose of glycyrrhizic acid. Conclusion: Piperine can inhibit the activation of cytochromes P450 remarkably, glycyrrhizic acid can relieving the inhibitory effect of piperine on liver cytochromes P450.
