摘要
目的观察还原型谷胱甘肽(GSH)合用粉防己碱(Tet)对帕金森病(PD)大鼠氧化应激水平的影响并探讨其作用机制。方法应用6-羟基多巴立体定向注射制作PD大鼠模型,将成功大鼠模型随机分为模型组、GSH组、GSH+Tet组、左旋多巴组,另设正常对照组,给药50d后测定鼠脑黑质线粒体酶复合体I活性及纹状体活性氧(R0S)含量。结果GSH+Tet对PD大鼠黑质线粒体酶复合体I有保护作用,GSH+Tet能明显降低PD大鼠纹状体活性氧(R0S)水平。结论GSH+Tet能减轻PD大鼠黑质及纹状体氧化应激损伤,并对线粒体呼吸链有一定保护作用。
Objective To observe the effects of glutathione(GSH) and tetrandrine(Tet) on nigral and striatum antioxidation system and mitochondrial complex I function in Parkinson disease (PD) model rat. Methods The PD rats model were established through stereotoxic microinjection of 6-OHDA.The model rats were randomly divided into 4 groups:model group, GSH group, GSH+ Tet group,L-dopa group and normal group. In each group drug treatment was given for 50 days. The activity of mitochondrial complexI in nigra and ROS in striatum were assayed after treat-ment. Results GSH+Tet group could increasely decrease the contents of ROS in striatum. GSH+ Tet could produce some protective effect on mitochondrial respiration chain. Conclusion GSH+Tet group could decrease the nigral and striatum oxidative stress in PD model rats,and produce some protective effect on mitochondria respiration chain.
