摘要 目的探索利用人成纤维细胞和York猪脱细胞真皮基质(ADM)微粒复合构建组织工程微粒皮肤的可行性并对其进行初步研究。方法体外培养取自胎背的人成纤维细胞,用PKH26标记,将细胞与ADM微粒复合构建微粒皮肤,用荧光显微镜观察、电镜观察和MTT法检测组织工程微粒皮肤的性能。结果脱细胞真皮基质微粒均匀柔软,弹性、韧性好,三维结构完整且排列规则。细胞与ADM微粒的复合情况良好,2h后开始黏附,2d后增殖明显,10d增殖达到高峰;7d时取材,HE和SEM显示成纤维细胞在ADM微粒表面成层黏附,并分泌大量基质;细胞生长良好。结论利用人成纤维细胞和York猪的ADM微粒可以构建具有活性的组织工程微粒皮肤。 Objective To explore the feasibility of construction the tissue engineering granule skin with human fibroblasts and York pig's acellular dermal matrix (ADM) granules. Methods The Human embryo fibroblasts were cultured in vitro and labeled with PKH26,then combined with ADM microgranules to construct granule skin. The characteristics of tissue engineering microgranular skin were measured with fluorescent microscope, SEM and MTF assay. Results The acellular dermal matrix granules were showed homogeneous soft with good elasticity and toughness,integrated 3D construction and regular array. The combination condition of cells and ADM microgranules was satisfactory, adhered in 2 hours, starting proliferation in 2 days and reach peak at 10 days. The samples adopted at 7 days showed the fibroblasts layed adhered at the ADM microgranular surface and well growing with abundant secreation of matrix. Conclusion It is demonstrated that the active tissue engineering microgranular skin might be constructed with human fibroblasts and ADM microgranules by York pig.
