摘要
目的探讨泰素帝合并PDTC对PC-3人前列腺癌细胞的杀伤效应及机制。方法应用流式细胞术、RT-PCR、免疫细胞化学方法,观察不同处理后PC-3细胞凋亡率及NF-κB表达的改变。结果PDTC组PC-3细胞凋亡率和死亡率分别为12%和6.8%,泰素帝组为12.5%和7.2%,两药合并后,凋亡率和死亡率分别增加为25.5%和11.5%;泰素帝有促进NF-κB表达和向核内转移作用;PDTC对NF-κB表达及核转位具有抑制作用;两者合并,NF-κB表达量及核转位减少,与单独PDTC相近。结论泰素帝与PDTC均能诱导PC-3细胞凋亡,并且两者具有协同作用,其机制可能与PDTC拮抗泰素帝的激活NF-κB效应有关。
Objective To study the lethal effect and the mechanisms of Docetacel combined with PDTC on human prostate cancer cell line PC-3, Methods The apoptosis rate and expression of NF-κB in PC-3 cell line were observed by means of flow cytometer, reverse transcriptase polymerase chain reaction (RT-PCR) and immunocytochemistry after different treatments. Results Apoptosis rate and death rate were 12% and 6.8% in PDTC group, 12.5% and 7, 2% in Docetaxel group, and 25.5% and ll. 5% in the combination group respectively. Docetaxel promoted the expression of NF-κB and the translocation of NF-κB into the nucleus while PDTC had the opposite effects. When docetaxel combined with PDTC, the expression and translocation of NF-κB were decreased, which were similar to using PDTC alone. Conclusion Both Docetaxel and PDTC can induce apoptosis in PC-3 cell line and they have synergistic effect. The mechanism maybe related with the fact that PDTC inhibits the Docetaxel-induced activation of NF-κB.
