摘要
拮抗放线菌RA-2发酵液经过乙酸乙酯萃取,浓缩,硅胶GF254薄层层析分离提纯到抗生素。对该抗生素进行不同温度、pH以及UV不同时间照射处理后,测定该抗生素的抑菌活性。试验结果表明:25℃水浴30min,该抗生素的抑菌活性为70.88%,在25 ̄65℃之间,抗生素的抑菌活性与原液的抑菌活性在P<0.05水平上无显著性差异。不同pH处理后,在pH5 ̄9之间,抗生素具有较强的稳定性。不同UV照射时间的处理后,照射15、45min的抑菌活性与未照射原液的抑菌活性在P<0.05水平上无显著性差异。拮抗放线菌RA-2所产的抗生素组分单一。硅胶GF薄层层析分离的组分2具有抑菌活性,为35.49%。
The antibiotics were isolated and purified by ehyl acetate extraction, concentration, thin layer chromatography through the fermentation broth of the antagonistic actinomycetes RA-2. The antibiotics were treated by different methods, including temperature, pH and radiating with ultraviolet, and its antifungal activity were determinated. The results showed that the antifungal activity of the antibiotics were 70.88% through 25℃water bath for 30min.The tested and without tested of antibiotics had no significant effect on the antifungal activity at the level P〈0.05 when it was at temperature 25-65℃. With different pH tested, the antibiotics had the better stability between 5and 9. Radiating with ultraviolet 15min and 45min, the antifungal activity had no significant effect on the antifungal activity at the level P〈0.05. The antibiotics had single fraction. Fraction2 through the TLC had the antifungal activity, 35.49%.
