摘要
①目的探索活性结膜的保存方法。②方法对深低温保存53,362,1123d的兔结膜,分别进行琥珀酸脱氢酶(SDH)、葡萄糖-6-磷酸脱氢酶(G-6-PD)、乳酸脱氢酶(LDH)、葡萄糖-6-磷酸酶(G-6-P)、腺苷三磷酸酶(ATP)活性测定;对深低温保存349~386d的兔结膜行同种异体移植实验,观察术后兔脾脏和结膜植片的组织病理学改变;对深低温保存76~535d的人尸结膜临床移植效果进行了随访观察。③结果深低温保存兔结膜的各种酶活性与对照组新鲜兔结膜无明显差异;术后8~78d,兔脾小结增生活跃,说明深低温保存兔结膜仍有抗原性,但结膜植片愈合良好,炎性细胞逐渐消退,仅上皮细胞层数减少。深低温保存人尸结膜临床移植后随访0.12~2.50年,全部结膜植片均原位存活,但移植30d后有的植片新生血管较多,有的植片呈轻度皱缩表现。④结论深低温保存结膜法是一种活性结膜保存法,可为临床随时提供移植材料,是眼库长期保存活结膜组织的一种较理想方法。
Objective In order to investigate the method of viable conjunctiva preservation,experimental and clinical researches on cryopreserved conjunctiva were carried out. Methods The enzymatic viabilities of SDH,G6PD,LDH,G6P and ATP of rabbit conjunctiva cryopreserved for 53,362 and 1123 days were determined .The experimental allografts using rabbit conjunctivae cryopreserved for 349 ̄386 days were performed and the histopathological changes of the rabbit spleens and conjunctiva grafts were observed after operation.The clinical transplantations with human cadaver conjunctivae cryopreserved for 76 ̄535 days were performed in 14 patients(14 eyes). Results The enzymatic viabilities of cryopreserved rabbit conjunctivae were resembling that of the control group of fresh rabbit conjunctivae .Eight to seventy eight days after operation , rabbit spleen nodules showed active hyperplasia , which suggested that cryopreserved conjunctivae still had antigenicity,but the conjunctiva grafts healed well and inflammatory cells disappeared gradually,only the number of epithlial cell layers decreased.All of the allografts kept survival during 1.5 months to 2.5 years followup with some grafts having vascularization and others a slight shrinkage one month after operation . These changes might be the result of rejection . Conclusion The cryopreservation method of conjunctivae is a viable conjunctiva preservation method. It is one of the ideal banking techniques for longterm preservation of viable conjunctivae and provide a conjunctiva sources available for clinical use at any time.
