基质金属蛋白酶-9与抑癌基因PTEN在大肠腺瘤、大肠癌组织中的表达及意义 被引量:9

摘要 目的:探讨MMP-9、PTEN在大肠癌、大肠腺瘤、大肠正常组织中的表达,进一步了解二者的关系.方法:通过免疫组化EliVision方法检测基质金属蛋白酶-9(MMP-9)与PTEN在肠癌组、腺瘤组及正常黏膜组中的表达,分析MMP-9与PTEN在大肠癌发病中的相关性.结果:正常组MMP-9的表达率明显低于肠癌组和腺瘤组(15.00%vs56.50%,45.00%,均P<0.01),正常组PTEN的表达率明显高于肠癌组和腺瘤组(95.00%vs39.10%,65.00%,均P<0.01),二者在腺瘤组与肠癌组间比较差别均无显著性.MMP-9在肠癌组中表达与肿瘤的分化程度呈负相关,与浸润深度、淋巴结转移呈正相关,PTEN蛋白表达与此相反,二者均与肿瘤的大小、患者的年龄均无相关性.MMP-9与PTEN在大肠癌中的表达具有相关性(r=-0.797,P<0.05).结论:MMP-9与PTEN在大肠癌发生发展中起重要作用,二者可作为评估大肠癌浸润转移的重要生物学指标. AIM: To investigate the expression ot matrix metalloproteinase-9 (MMP-9), PTEN in colorectal carcinoma, adenoma and normal mucosa of large intestine as well as their correlations. METHODS: EliVision immunohistochemical method was used to examine the expression of MMP-9 and PTEN in colorectal carcinoma, ade- noma and normal mucosa, and their correlations were also analyzed. RESULTS: The expression of MMP-9 was significantly lower in normal mucosa than that in carcinoma and adenoma (15.00% vs 56.50%, 45.00%;both P 〈 0.01), while the expression of PTEN was significantly higher (95.00% vs 39.10%, 65.00%; both P 〈 0.01). MMP-9 and PTEN expression were not significantly different between adenoma and carcinoma. MMP-9 expression was positively correlated with histological classification, but negatively correlated with lymph node metastasis and the depth of invasion. PTEN expression was just the opposite. Both MMP-9 and PTEN expression had no correlations with the age of patients and tumor size. There existed a significant correlation between the expressions of MMP-9 and PTEN in colorectal carcinoma (r =-0.797, P 〈 0.05). CONCLUSION: The expression of MMP-9 and PTEN may play important roles in the occurrence and development of colorectal carcinoma. MMP-9 and PTEN may serve as new markers in the diagnosis and prognosis of colorectal carcinoma.
