摘要
本研究旨在观察L-精氨酸对高胆固醇血症鹌鹑主动脉粥样硬化形成的抑制作用。取雄性日本鹌鹑30只,分为3组,每组10只。正常对照组喂基础饲料,高脂组喂1%胆固醇+3%猪油(高脂饲料);L-精氨酸组喂高脂饲料+饮水补充225%L-精氨酸。8周后处死动物,取出主动脉,形态学观察动脉粥样硬化病变和计算斑块面积百分比;获取血清作生化分析。结果:(1)高胆固醇血症诱导主动脉内膜增生及粥样硬化病变。(2)补充2.25%L-精氨酸,血清精氨酸浓度上升一倍。(3)L-精氨酸组主动脉斑块面积及内膜厚度与胆固醇组比较,分别减少34%和38%(P<0.01);主动脉壁、血清总胆固醇水平分别下降51%和33%(P<0.01);丙二醛下降56%(P<0.01);超氧化物歧化酶活性上升一倍(P<0.01)。这些结果提示,L-精氨酸可抑制高胆固醇血症诱导的鹌鹑主动脉粥样硬化病变形成,其机制可能涉及降脂和抗脂质过氧化损伤作用。
This article was to determine whether oral administration of L-arginine to hypercholesterolemic quails would inhibit atherogenesis. 30 male Japanese quails were randomized into three groups,each with 10 animals. Control group was fed a normal chow. High lipid group received atherogenic diets containing 1 % cholesterol and 3% fat. L-Arg group received atherogenic diets supplemented with 2. 25% L-arginine in drinking water. After 8 weeks, all animals were killed, the aortae were horvested for histo morphological study as well as for determination of atherosclerotic plaque surface area,intimal thickness and content of total cholesterol. Serum was obtained for biochemical analysis. The result showed that hypercholesterolemia induced aortal atherosclerotic lesions and intimal hyperplasia. The addition of 2. 25% L-arginine doubled the serum arginine concentrations. There were a 34% reduction in lesion suface area and a 38% redution in the intinal thickness in L-Arg group compared to that in high lipid group(P<0.05);totalcholesterol in serum reduced 51 %; malondialdehyde 56%; activity of superoxide dismutase increased 54%. our study suggested that oral L-arginine might reduce the severity of aorticatherosclerotic lesions in hypercholesterolemic quails. The mechanisms of antiatherogenic effects of L-arginine in the hyper cholesterolemic quails might involved in its lowering blood lipids and reducing the injuries caused by lipid perorides.
