性激素拮抗剂对雄性大鼠血脂代谢的影响 被引量:2

The effects of androgen and estradiol antagonists on lipid metabolism in male rats

摘要 目的探讨雌、雄性激素拮抗剂对雄性大鼠血脂代谢的影响。方法18只雄性SD大鼠,随机分为对照组、雄激素拮抗组和雌激素拮抗组,每组6只,各组饲喂高脂饮食。对照组和雄激素拮抗组、雌激素拮抗组分别用等量生理盐水和氟他胺溶液(1mg·kg-1·d-1)、他莫西芬溶液灌胃(2mg·kg-1.d-1)。干预处理14w后,处死全部实验动物,心脏取血离心后,酶法测定血浆血脂相关指标,放免法检测血清睾酮、雌二醇水平。结果雌激素拮抗组血浆总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)水平低于对照组(P<0.05),高密度脂蛋白胆固醇(HDL-C)水平高于对照组(P<0.05);雄激素拮抗组:血浆TC水平高于对照组(P<0.05);血浆HDL-C、LDL-C水平与正常对照组之间无统计学差异;各组间血浆甘油三酯(TG)水平无统计学差异(P>0.05)。结论在雄性大鼠,内源性雄激素可以起到降低TC,保护心血管系统的作用;内源性雌激素可能升高血脂的相关指标,表现出对雄性大鼠心血管系统疾病发生发展不利的一面。 Objective To observe the effect of endogenous androgen and estradiol antagonist on serum lipids in high-fat diet male rats. Methods 18 male SD rats were randomly divided into antiandrogen, antiestrogen and control groups, 6 rats in each group. The rats in all groups were given with high fat diet. Control, antiandrogen and antientrogen groups were respectively intragastricly administrated with the same quantity sodium chloride, flutamide ( 1 mg·kg^-1·d^-1 ), and tamoxifen (2 mg·kg^-1·d^-1). All rats were killed 14 weeks later to obtain and centrifugate blood samples. Total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) concentrations in serum were measured by e.nzymatic technique. Serum testosterone and estradiol levels were detected by radioimmunity. Stastical analysis was done by one-factor analysis of variance. Results TC and LDL-C were lower ( P 〈 0. 05 ), but serum TG and HDL-C were higher in antiestrogen group than those in control group ( P 〈 0. 05 ). The levels of serum TC in antiandrogen group were higher than that in control group ( P 〈 0. 05 ), however there was no significant difference in HDL-C, LDL-C between antiandrogen and control groups ( P 〉 0.05 ). Conclusions lnhibiton of endogenous androgen could increase the level of serum TC, indicating that it could decrease TC to protect male rats' cardiovascular system. But inhibition of endogenous estrogen could decrease serum TC and serum LDL-C, indicating it could increase blood lipid related indexes to damage male rats' cardiovascular system.
