摘要
目的:观察体外分离培养的骨髓基质细胞(marrowstromalcells,MSCs)与温敏型水凝胶壳聚糖/甘油磷酸钠(chitosan/glycerophosphate,C/GP)的生物相容性。方法:实验于2006-03/12在白求恩国际和平医院实验室完成。SD大鼠MSCs经过体外培养传代后,与制备温敏型水凝胶C/GP在培养板内共培养7d,同时设立细胞对照组(仅加入细胞培养基)。采用形态学观察、MTT法检测MSCs在C/GP的生长情况,并绘制生长曲线。结果:①MSCs接种于C/GP复合物24h后见细胞成球状,未见明显梭形及多角形细胞,48h内生长速度较慢,72h后可见梭形、多角形细胞,细胞数量明显增多。②绘制MSCs生长曲线为“S”形,第1个24h为细胞潜伏适应期,第2~3个24h为对数生长期,4d后细胞增殖减慢,5d后细胞基本停止生长。细胞对照组的细胞潜伏适应期为第1~2个24h,对数生长期推后至3~5个24h,1周后细胞基本停止生长。结论:MSCs在C/GP凝胶中可良好存活、增殖,MSCs与C/GP凝胶有良好的生物相容性。
AIM: To explore the biocompatibility of thermosensitive chitosan/glycerophosphate (C/GP) gelatum and marrow stromal cells (MSCs) cultured in vitro.
METHODS: The experiment was performed at the laboratory of Bethune International Peace Hospital from March to December 2006. SD rat-derived MSCs were passaged in vitro and cultured in culture plate with C/GP for 7 days. In addition, cells cultured with culture medium only served as controls. Cell growth was observed by morphology and growth curve of MSCs was drawn using MTT method.
RESULTS: ①MSCs cultured in C/GP were globular after 24 hours of culture with C/GP. No obvious spindle or polygon-shaped cells were found. Cell grew slowly in 48 hours, and spindle and polygon-shaped cells were observed after 72 hours. ②The growth curves of MSCs in C/GP were shown as a sigmoidal shape. The first 24 hours was the adaptation period of cells; the second and third 24 hours was the log phase growth phase. Cell growth retarded after 4 days, even stopped on the 5th day. For control cells, the first and second 24 hours was the adaptation period, and the third and fifth 24 hours was the log phase growth. Cell growth stopped in one week.
CONCLUSION: MSCs survive and proliferate well in C/GP. The biocompatibility of MSCs to C/GP is very good.
