摘要
本研究以~3H哌唑嗪为特异的标记配基定量地检测了家兔心脏α_1受体及其在心脏缺血/再灌注损伤时的变化,并结合细胞膜内Ca^(2+)含量的变化及丹参对缺血心肌的保护作用,探讨了心肌/再灌注损伤的机理。结果表明,α_1受体随心肌缺血时间的延长而明显升高,40分钟时达正常对照的2倍以上,在此基础上再灌注4分钟,受体数目稍降,但仍保持在高水平。细胞膜的Ca^(2+)在缺血时的变化与α_1受体的变化平行(随时间而增高),但缺血40分钟再灌注4分钟时明显下降。预先耳缘静脉注射丹参制剂后,缺血40分钟再灌注4分钟,上述两种指标均与对照组无差异。提示丹参对心肌细胞膜具有明显保护作用。
We used radicligand binding assay to determine the changes of α_1-receptors in membrane of the myocardial cell during ischemia and reperfusion. The content of Ca^+ were measured by atomic absorption method. Bmax of α_1-receptors were increased remarkably after 30 min of myocardial ischemia and persisted for at least 40 min and 4 min(the time of ischemia and reperfusion)and recovered from 20 min of reperfusion, The Ca^(2+) content were increased significantly from 30 min to 40 min of ischemia and then quickly recovered(after 4 min of repefusion).
