摘要
目的:了解环一磷酸腺苷(cAMP)、环一磷酸鸟苷(cGMP)含量的变化与脑缺血、再灌注损伤和功能恢复间的关系。方法:以大鼠四血管阻断为模型,采用放免法测定不同状态下血浆和皮层、海马、纹状体中cAMP、cGMP的含量。结果:cAMP在各脑区和血浆中的含量,缺血组、再灌组、腺苷和N6环乙烷基腺苷(CHA)给药组分别与对照组相比均无显著性变化(P>005)。cGMP的血浆含量由对照组的532±135pmol/ml增高至再灌组的1846±417pmol/ml(P<001)。给药3、4组也分别增高至2015±562pmol/ml和2265±834pmol/ml,(P<001),但与再灌组相比无显著性差异(P>005)。其余血浆各组及脑区各组cGMP含量均无显著性变化。结论:各脑区内cAMP和cGMP含量与大鼠脑缺血损伤和恢复无直接关系。血浆内cAMP含量无显著性变化,cGMP含量虽有变化,但作为临床观测指标证据不足。
Objective:To look into the relationship between concentrations of cAMP and cGMP and cerebral ischemia,reperfusion damage and the recovery by adenosine treatment.Methods:Four vessel occlusion model of forebrain ischemia in rat was used. Since cerebral cortex、hippocampus and striatum are the most vulnerable areas to ischemic insult,the concentration of cAMP、cGMP in these areas and plasma were determined by RIA method. Results:The concentration of cAMP in either plasma or different cerebral areas was not changed neither in ischemic group、reperfusion group nor in treatment group with adenosine and N 6 cyclohexyladenosine(CHA),an adenosine receptor agonist,compared with that of control group. The concentration of cGMP in plasma increased from 5 32±1 35 pmol/ml in control group to 18 46±4 17 pmol/ml in reperfusion group,( P <0 01 vs CONT). In treatment 3 and 4 groups it also increased to 20 15±5 62 pmol/ml and 22 65±8 34 pmol/ml,respectively,( P <0 01 vs CONT).The concentration of cGMP of other groups and all the groups in different cerebral areas was not changed.Conclusions:The concentration of cAMP and cGMP in different cerebral areas has no direct relationship with the damage of cerebral ischemia and resuscitation.Although the concentrations of cGMP in plasma were changed,there are not enough evidence to use it as a clinical index.
