沙土鼠脑缺血预处理对缺血再灌注后神经细胞凋亡的影响

摘要 目的探讨缺血预处理对沙土鼠脑缺血再灌注后神经细胞凋亡的影响。方法采用阻断双侧颈总动脉制作沙土鼠全脑缺血模型,96只沙土鼠,腹腔注射戊巴比妥钠麻醉。沙土鼠随机分为四组:SH组:假手术对照组,未行脑缺血预处理;IC组:预处理对照组,予2 min脑缺血后未再次缺血;IP组:预处理缺血组,予2 min脑缺血,再灌注24 h后,再次脑缺血10 min;IR组:脑缺血再灌注组,予脑缺血10 min后行再灌注。每组据再灌时间点的不同又分为Ⅰ(1d)、Ⅱ(3d)、Ⅲ(5d)、Ⅳ(7d)4个亚组,每个亚组6只动物。采用TUNEL法检测沙土鼠脑海马CA1区神经细胞凋亡数目。结果IP可显著减少沙土鼠脑海马CA1区神经细胞凋亡数量(与IR组相比P<0.01)。结论脑缺血预处理可显著提高机体对后续缺血的耐受性,其机制可能是通过抑制缺血诱导的神经细胞凋亡而发挥脑保护作用。 Objective: To investigate the influence of the cerebral ischemic preconditioning on the neural apoptosis following ischemia and repeffusion in gerbils, Methods- Cerebral ischemia was induced by occlusion of bilateral common carotid arteries. Gerbils were randomly divided into four groups- Sham- operation control group (SH) : without cerebral ischemia; Cerebral ischemic preconditioning control group (IC) ; 2 rain cerebral ischemia only; Cerebral ischemic preconditioning test group (IP) - 2 min cerebral ischemia and 24 h reperfusion followed by cerebral ischemia 10 min; And cerebral ischemia - reperfusion (without preconditioning) group (IR). Observations were carried out in each subgroup 1 d、3d, 5d and 7d (each subgroup n =6), the apoptosis neurons in hippocampus CA1 region was counted. Results: The number of apoptosis neurons in hippocampus CA1 region were much less in group IP than in group IR ( P 〈 0.01 ). Conclusion: Ischemic preconditioning can significantly enhance the tolerance of the following ischernia, The mechanism of protection may be that ischemic preconditioning can prevent neurons from apoptosis induced by ischemia.
