摘要
目的:研究内皮素-1对肾小球系膜细胞周期G1期进展的调控机制。方法:应用免疫组织化学、免疫印迹分析等技术检测系膜细胞在内皮素-1刺激增殖时,细胞内G1期调控蛋白cyclinD1和cdk4表达的变化。结果:正常指数式生长的系股细胞核内表达二种蛋白,在内皮素1刺激下G1期进展中cyclinD1表达增加,cdk4表达变化不明显。结论:cyclinD1是体外培养肾小球系膜细胞针对炎症因子内皮素1刺激时G1期进展的重要调节蛋白。
Objective: To investigate the mechanism of G1 progression in rat mesangial cells responded to Endothelin-1. Methods: Immunohistochemical stainning and Western blotting analyses were used to detect the changes of G1 regulators Cyclin D1 and cdk4 protein in mesangial cells stimulated by Endothelin-1. Results:Cyclin D1 and cdk4 were expressed located in the nuclus of exponentially growing rat mesangial cells, and the level of cyclin D1 protein increased during G1 progressing, but that of cdk4 did not change prominently.Conclusion: Cyclin D1 may act as an important regulator of G1 progession in rat mesangial cells responded to Endothelin-1 stimualtion.
