摘要
目的研究中药癃必消胶囊对体外培养的人前列腺增生间质细胞Fibronectin和Collagen IV基因表达的影响。方法把含癃必消胶囊的药物血清加入到体外培养的人前列腺增生间质细胞中、采用实时定量RT-PCR等相关技术,检测Fibronectin和Collagen IV在体外培养的人前列腺增生间质细胞中的表达。结果癃必消胶囊能降低体外培养的前列腺增生间质细胞Collagen IV基因的表达,对Fibronectin基因的表达无明显规律。结论癃必消胶囊通过抑制前列腺间质细胞Collagen IV基因的表达达到治疗前列腺增生的目的。
Objective To investigate the effects of Chinese herbal medicine Long-Bi-Xiao （LBX）capsule on Fibronectin and Collagen Ⅳ expression in human prostatic stromal cells cultured in vitro. Methods Stromal cells were isolated and cultured from the human prostatic stroma and subcultured. Normal rabbits were medicated with LBX and their blood serum were collected to prepare the medicated serum, which was incubated with the subcultured stromal cells, Real-time RT=PCR was applied to detect the mRNA expression levels of Fibronectin and Collagen Ⅳ. Results LBX reduced the expression of extracellular matrix collagen Ⅳ, but its effect on fibronectin expression was not clear. Conclusion LBX treated BPH through reducing stromal cell collagen Ⅳ mRNA expression.
