银杏花粉微管蛋白的纯化及鉴定 被引量:3

Purification and Identification of Tubulin from Ginkgn biloba Pollen

摘要 采用丙酮粉抽提,DEAE-SephadexA-50、SephacrylS-300、MonoQ柱层析,从银杏(GinkgobilobaL.)花粉中分高纯化出微管蛋白(tubulin),其两个亚基(α、β)的分子量分别为54kD和52kD.纯化的微管蛋白可与鸡脑微管蛋白抗体发生免疫交叉反应. Chnkg lelde is a ooical gyYnnOSPe and occupies an important P0Stbo in the evo-lution 0f plant kingd0m. Tubulin was purified from ginkg POlen by acetone preciPitatio, fol-lDwd by DEAE-SePhadex A5O chr0rnatograPhy, SePhaCry S3OO gel filtration, and M0nO Q co-uxnn chrOrnatogrphy. The purified tubulin is electmphoretically hanOgeneO and is cornPOSed 0ftwo shubunits. The aPParen molecthar wewhts of the two subunits are ahaut 54 and 52 ari re-sPectivey, estirnated han the StoPAG.
