摘要
以胸苷为原料,先用甲磺酰氯将3′,5′-羟基保护制得化合物(2),然后在氢氧化钠水溶液中形成3′,5′-O-环状物(3),再与叔丁醇钾作用生成2′,3′-二脱氧-2′,3′-二脱氢胸苷(4),最后通过Pd-C加氢得到2′,3′-二脱氧胸苷(5),总收率为33.6%.
The 2′,3′ dideoxythymidine posseses antiviral activity.From 2′ deoxythymidine,3′,5′ di O mesylthymidine(2) was prepared in 86 3% yield,which in treatment with NaOH in H 2O gave 67 4%(3),then treated with KOBu tert in DMSO to give 90%(4).Finally,(4) reacted with H 2 to yield 64 1%(5).The total yield of (5) was 33 6%.
