rhBMP2对牙髓成纤维细胞增殖及碱性磷酸酶活性的影响 被引量:6

摘要 目的了解重组人骨形成蛋白(rhBMP2)对牙髓成纤维细胞增殖活性及碱性磷酸酶(ALP)活性的影响。方法用MMT比色法检测不同浓度rhBMP2作用下,牙髓成纤维细胞增殖活性的变化;用酶动力学方法检测不同浓度rhBMP2作用下ALP活性的变化。结果低浓度的rhBMP2可提高体外培养人牙髓细胞的增殖活性,浓度为200ng/mL时,增殖活性达到最大值,与对照组间差异有统计学意义(P<0.05)。并可促进人牙髓细胞ALP活性,呈浓度依赖性,与对照组间差异有统计学意义(P<0.05)。结论rhBMP2能够促进牙髓细胞增殖和ALP活性,是牙髓细胞增殖和分化的重要因子之一。 Objective To know the effect of recombinant human bone morphogenetic proteins -2 (rhBMP2)on proliferation of dental pulp fibroblast and alkaline phosphatase(ALP)activity. Methods MTT method and enzyme kinetics method were respective ly used to determine the cell proliferation and ALP activity under different concentration of rhBMP2. Results rhBMP2 at the low concentration could increase the proliferation of dental pulp fibroblast in vitro . The group with 200 ng/mL rhBMP2 showed the best activity of proliferation and there was statistic difference between test group and control group(P〈0.05). ALP activity was gradually increased with the increasing of rhBMP2 and there were statistic differences between groups(P〈0.05). Conclusion rhBMP2 is an important factor promoting the proliferation and differentiation of dental pulp fibroblast,which can increase the proliferation of dental pulp fibroblast and activity of ALP.
