培养的心内膜内皮细胞形态和旁分泌特点的初步研究

摘要 心内膜内皮(EEC)是心脏功能重要的调节者。本实验观察培养的新生小牛右心室EEC形态特点及其血管活性物质的分泌功能。培养的EEC较同样条件下培养的肺动脉内皮细胞(PAEC)大而伸展。免疫组化发现EEC的VIII因子相关抗原(VIII_(Ag))、心钠素(ANP)染色较弱,而EEC的内皮素(ET-1)阳性反应染色较PAEC强。采用放免测定发现EEC可向培养液中释放分泌ET-1,其释放量是PAEC的2.6倍,其血管紧张素Ⅱ(ATⅡ)分泌量与PAEC类似,但ANP和内源性洋地黄因子(EDLF)的释放量显著低于PAEC(P<0.01)。上述观察揭示,右心室EEC能通过旁分泌血管活性成份特别是能分泌大量的ET-1,从而调节心肌功能,同时对肺循环也可能起重要的调节作用。 The endocardial endothelium modulates the performance of the subjacent myocardium In the present study, 'morphological and paracrine characteristics of cultured bovine endocardial endothelial celis (EEC) in vitro were investigated and compared with pulmonary artery endo-thelial celis (PAEC)harvested from the same animals and cultured under the same conditions. Morphologically, EECs were much larger and flatter than PAECs. Immunochemistry demonstra-ted more positive endothelin-1 (ET-1) staining, less positive atrial natriuretic factor (ANP) and factor Ⅷ related antigen staining in EEC than in PAEC. The rate of ET-1 release was 2.6-fold (P<0.01) greater, whereas that of ANP and endogenous digitalis-like factor (EDLF) were lower from EEC than from PAEC (P<0.01), that of ahgiotensin Ⅰ (AT Ⅱ) release was similar. The above results suggest EEC is different from PAEC and a greater source of ET-1.The paracrine of vasoactive factors by EEC may play important roles in regulating car-diac function and pulmonary circulation.
